Items

ESP-Bruce at WHOI

  •  We will have Bruce K. remove temp-sensitive reagents and FedEx to us, and we will ship equipment back in Unrefrigerated van ($5800 vs $9200).  Because of Keafer's schedule, ESP will not be shipped until July 25.  Roman sending Hobo temp sensor to see what temp swings the van experiences.
  • We need to spot-check the ESP temp calibrations upon return, just to be sure.

Pucks

  •  We have had 1200 puck pieces in need of serial number etching.  We've rec'd the first 400 back from the etcher.  Nilo is inspecting and will ready the next 800, probably in batches of 400.
  • We've also rec'd pucks owed us from McLane (already serialized).  These need to be inspected and Roman planning on doing lysis test with them.

CTD

  •   Newly rebuilt fluorometer seems to give same readings as an old working model.  Roman/Nilo doing an 'at-sea' test right now--I'm watching them out the window bounce around in the Whaler.
  • We need to know the range the fluorometer needs for the clean water at Station Aloha.  It may need 100x sensitivity.  So, order of events:
    1. Confirm CTD-4894 is working
    2. Call Kilo Muana for their CTD sensitivity settings
    3. Call/send to Seabird to make the change
  • Once drifter cruise is over, we will need to lower the gain for 'normal' use again.  Next use may be the CA current drift in the Fall, 2011.

Drifter Cruise

  • We have the Shana Rae lined up for at-sea test either week of Aug8 or Aug15.
  • Next Tuesday (Jul 19) we will meet with Julie & Roman to start intense logistical planning.
  • Chris would like to meet with Julie later in week to discuss:
      • All data collected so far on MFB related to drifter preparations
      • Progress on ISME paper
  • Roman has array printing for this deployment completely under control.

Sybr HAB Assay development

  •  at 400nm, results on ABI looked good.  MFB look similar to AOAb.  Raised primer concentration and got much better signal on MFB.
  • Elif rec'd many cultures of various pseud's and is making plasmids, as these cultures have become difficult to get.

SC Wharf Deployment

  • We will start the ball rolling on this now.  Looking at a mid/late October deployment.
  • Should check with Ralphe about his CTD and the wharf guys about installing a new pump (Sept.)
  • Elif would like to continue helping the QC of arrays, so it's not all up to one person (Chris P has Aug vacation, as does Kevan in Sept.)

Other Items

  • We will decide by July 22 whether we will participate in the CANON Sept cruise.  Things to learn by then to help in decision:
      • what would be the mission?
      • Are we chasing upwellings or is it HAB related?
      • repeat of Spring deployment with adaptive sampling?  Yanwu would need to be involved.
